Application:
The growth of breast cancer cells depends on the presence of estrogen, and the estrogens (estrone and estradiol) circulating in postmenopausal women are mainly converted by aromatic enzymes in the peripheral tissues to androgens (androstenedione and testosterone) in the adrenal glands and ovaries. Inhibiting estrogen production by inhibiting aromatase is an effective selective treatment for postmenopausal hormone-dependent breast cancer. Exemestane is a kind of irreversible steroidal aromatase inactivator, the structure and nature of the enzyme substrate androstenedione, similar to the pseudo substrate of aromatase, through irreversible to combine with the enzyme's active site and make its inactivation (the role is also referred to as "self-destruct inhibition"), which significantly reduce postmenopausal estrogen levels in the blood circulation, but in adrenal cortical steroids and aldehyde sterol biosynthesis has no obvious influence. Other enzymes in the steroidogenic pathway were not significantly affected when the concentration of inhibition of aromatase action was 600 times higher than that of inhibition.
