Pharmacological effects:
Thianaputin sodium is a tricyclic antidepressant. Its antidepressant mechanism is different from that of traditional tricyclic antidepressants. It can increase the reuptake of 5-ht in the synaptic gap and may have the effect of improving the neuronal conduction of 5-ht. No affinity for choline or adrenaline receptors. Antidepressant efficacy was similar to tricyclic, but better tolerated than tricyclic. In animal experiments, it can increase the spontaneous activity of pyramidal cells in the hippocampus and accelerate the recovery of their functions after inhibition. Increased serotonin reuptake by neurons in the cerebral cortex and hippocampus. Thianapur sodium is absorbed rapidly and completely in the digestive tract. The distribution was rapid, with a PPB of up to 94%. The metabolism of the liver is complete through the hypo-oxidation and n-demethylation, and the terminal t1/2 is short, which is 2.5h. A very small amount of the protodrug (8%) is excreted from the kidneys, mainly its metabolites from the urine. Clearance of t1/2 was prolonged for 1h in patients with renal insufficiency.
Indications:
It is used to treat mild, moderate or major depression, neurogenic and reactive depression, anxiety depression with somatic, especially gastrointestinal distress, and anxiety depression in alcohol-dependent patients during withdrawal.
