Pharmacological effects:
1. Pharmacology ketoconazole is a pyrrole antifungal agent, which has antibacterial effects on deeply infected fungi, such as candida, chromatitis, coccidioides, histoplasmosis, spore filariae, etc., and also has antibacterial activities against tinea trichoderma. This product has weak effect on aspergillus, schenkella, some melanosporaceae, trichoderma, etc. By interfering with the activity of cytochrome p-450, this product inhibits the biosynthesis of ergosterol, the main sterol in the cell membrane of fungi, damages the cell membrane of fungi and changes its permeability, resulting in leakage of important intracellular substances. Ketoconazole can inhibit the biosynthesis of triacylglycerol and phospholipid of fungi, inhibit the activity of oxidase and peroxidase, and cause the accumulation of hydrogen peroxide in cells, leading to cell submicrostructure degeneration and cell necrosis. Candida albicans inhibited the transition from spores to invasive hyphae. Toxicology animal long-term toxicity experiments show that ketoconazole can significantly increase alkaline phosphatase, hepatocyte degeneration.
Indications:
Ketoconazole is suitable for the treatment of the following systemic fungal infections: 1. Candidiasis, chronic cutaneous and mucosal candidiasis, oral candidiasis, urinary tract candidiasis, chronic and recurrent vaginal candidiasis with no local treatment. 2. Dermatitis blastomycosis. 3. Coccidioidomycosis. 4. Histoplasmosis. 5. Pigmented mycosis. 6. Paracoccidiosis. Dermatomycosis, tinea versicolor and tinea tinea caused by dermatophytes and yeasts can be treated with this product when local treatment or oral administration of griseofulvin is ineffective, or when severe intractable fungal skin infections cannot be treated with griseofulvin.
