Application:

Studies in vivo and in vitro show that letrozole can effectively inhibit the transformation of androgens to estrogen, and the estrogen of postmenopausal women is mainly derived from the precursor substance of androgens in the peripheral tissue aromatation, so it is particularly suitable for postmenopausal breast cancer patients. Letrozole was 150 ~ 250 times more active than the first generation aromatase inhibitor aminorumit. Due to its high selectivity, it does not affect glucocorticoids, saline corticosteroids and thyroid function, and large doses of the drug have no inhibitory effect on the secretion of adrenocorticosteroids, so it has a high therapeutic index. Various preclinical studies have shown that letrozole has no potential toxicity to all systems and target organs of the body, and has good tolerance and strong pharmacological effects. Compared with other aromatase inhibitors and anti-estrogens, letrozole is more effective against cancer.

Indications:

1. Treatment of advanced postmenopausal breast cancer (estrogen receptor, progesterone receptor positive or unknown receptor status), mostly used for second-line treatment after failure of anti-estrogen therapy;

2. First-line treatment for locally advanced or diffuse postmenopausal breast cancer;

3. Extended adjuvant therapy for postmenopausal breast cancer patients who had received standard adjuvant tamoxifen therapy for 5 years;

4. It is used for postoperative adjuvant therapy for hormone-positive early breast cancer patients.
