Megestrol is a 17 benzodiazepine-hydroxyprogesterone derivative, which is a highly effective synthetic progesterone. Its progesterone effect is about 75 times that of progesterone when taken orally and 50 times that of progesterone when injected. This product has obvious anti-estrogen effect, no estrogen or androgen activity, no protein assimilation effect. It can inhibit the release of hypothalamic gonadotropin-releasing hormone (GnRH) and be used for adenohypophysis to reduce its sensitivity to GnRH, thereby blocking the release of pituitary gonadotropin and producing significant ovulation inhibition. Therefore, it is used in combination with ethinylestradiol as a short-acting contraceptive. This product can also make cervical mucus become sticky and thick when used in large doses alone, which is not conducive to the passage of sperm, and inhibits the normal development of endometrial glands, preventing the implantation of fertilized eggs.

In addition, this product has a certain inhibitory effect on hormone-dependent tumors. The mechanism may be to control the development and growth of ovarian follicles through the effect on the secretion of pituitary gonadotropin (FSH), so as to reduce the production of estrogen. In addition, it can also act on estrogen receptors, interfere with the combination of estrogen and inhibit the growth of tumor cells.

Indications:

Mainly used as a short-acting oral contraceptive, but also as an intramuscular injection of long-acting contraceptive. It is also used to treat dysmenorrhea, amenorrhea, functional uterine bleeding, endometriosis and endometrial adenocarcinoma.
