Method name:
Determination of tetracaine hydrochloride - neutralization titration method
Application:
This method used titration method to determine the content of tetracaine hydrochloride.
This method is suitable for tetracaine hydrochloride.
Method principle:
After adding glacial acetic acid and acetic anhydride test solution, the test was heated and reflux for 2 hours, then cooled down. After adding mercury acetate test solution and crystal violet indicator solution, the solution was titrated with perchloric acid titrant (0.1mol/L) until the solution turned blue. The amount of perchloric acid titrant was recorded and calculated.
Reagents:
1. Water (let boil to room temperature)
2. Perchloric acid titrant (0.1mol/L)
3. Crystal violet indicator
4. Acetic anhydride
5. Glacial acetic acid
6. Mercury acetate test solution
7. Potassium hydrogen phthalate as reference
Equipment:
Sample preparation:
1. Perchloric acid titrant (0.1mol/L)
Preparation: take anhydrous glacial acetic acid (calculated according to water content, add acetic anhydride 5.22ml for every 1g of water)750mL, add perchloric acid (70%-72%) 8.5ml, shake well, slowly add acetic anhydride 23mL at room temperature, shake while adding, shake evenly after adding, let cool, add anhydrous glacial acetic acid right amount to make 1000mL, shake well, leave for 24 hours. If the test product is easy to acetylate, the water content of this page must be determined by water content determination method, then water and acetic anhydride to adjust the water content of the solution is 0.01%-0.2%.
Calibration: the reference potassium hydrogen phthalate dried to constant weight at 105℃ was 0.16g, then it was precisely weighed, dissolved by adding 20mL of anhydrite acetic acid, and 1 drop of crystal violet indicator solution. The titration was titrated to blue slowly, and the titration results were corrected by blank test. Each 1mL perchloric acid titrant (0.1mol/L) is equivalent to 20.42mg of potassium hydrogen phthalate. According to the consumption of the liquid and the amount of potassium hydrogen phthalate, the concentration of the liquid can be calculated.
Storage: store in brown glass bottle and keep closed.
2. Crystal violet indicator
Take 0.5g crystal violet and add 100mL glacial acetic acid to dissolve it.
Operation steps:
Precision according to 0.2 g of the test, add 5 mL of 25 mL glacial acetic acid and acetic anhydride dissolves, heating reflux 2 hours, put the cold, add acetic acid liquid mercury try 5 mL and indicating liquid crystal violet 1 drop, with perchloric acid titration fluid (0.1 mol/L) blue titration solution, and use the result of the titration blank test correction, record consumption of perchloric acid titration fluid volume (mL), each 1 mL perchloric acid titration fluid (0.1 mol/L) is equivalent to 30.08 mg of tetracaine hydrochloride (C15H24N2O2 · HCl), a quick.
